How it works
MK-677 activates the ghrelin receptor. That has two main effects:
- More growth hormone, released in natural pulses by the pituitary, which raises IGF-1. In trials, daily use raised IGF-1 for as long as it was taken.
- More hunger, because ghrelin is the body’s main appetite signal.
Its half-life is about 24 hours, so it’s taken once a day. Studies show increases in lean mass, but not consistent gains in strength or function.

Blood sugar
